Can I run Zune software from an External HD?

I downloaded the Zune software from the net, but onto my external. Everything looks to be in order, but when I click to start it (from the desktop shortcut or the external), it tells me it can’t run until I re-start my computer. I’ve done this several times. With and without the external plugged in. Any ideas??
